2020 - Czech The aim of the methodology is to acquaint the professional public and users with the information obtained and the evaluation of basic methods of mucus collection in fish. Based on the knowledge and evaluation, to enable the most efficient collection of material for subsequent research. The target group of the methodology are mainly persons from veterinary practice and ichthyologists dealing with the issue of fish integument, both in relation to their health status or developmental-anatomical studies. Representatives of different taxonomic groups were preferred in the selection of fish species for verification. Cílem metodiky je seznámit odbornou veřejnost a uživatele se získanými informacemi a hodnocením základních způsobů odběru hlenu u ryb. Na základě poznatků a hodnocení tak umožnit co nejefektivnější odběr materiálu pro následný výzkum. Cílovou skupinou dané metodiky jsou zejména osoby z veterinární praxe a ichtyologové zabývající se problematikou integumentu ryb, a to jak ve vztahu k jejich zdravotnímu stavu nebo vývojově-anatomickým studiím. Při výběru druhů ryb k ověření byly preferovány zástupci různých taxonomických skupin. Methodology for measuring the character of the mucus layer in fish

2020 - Czech The aim of the methodology is to enable easy quantification of selected characteristics of the fish integument, namely the number of secretory cells in the surface layer of the skin, the amount of epidermal melanophores and the ability of the mucus layer to absorb UV radiation. The target group of the methodology are mainly persons from veterinary practice and ichthyologists dealing with the issue of fish integument, in relation to their health status or developmental-morphological studies. It can also be used in fish farming or protection. Representatives of different taxonomic groups were preferred in the selection of fish species for verification. Cílem metodiky je umožnit snadnou kvantifikaci vybraných charakteristik rybího integumentu, konkrétně množství sekrečních buněk v povrchové vrstvě pokožky, množství epidermálních melanoforů a schopnost hlenové vrstvy absorbovat UV záření. Cílovou skupinou metodiky jsou zejména osoby z veterinární praxe a ichtyologové zabývající se problematikou integumentu ryb, a to ve vztahu k jejich zdravotnímu stavu nebo vývojově-morfologickým studiím. Uplatnění může dále nalézt i v rámci chovu či ochrany ryb. Při výběru druhů ryb k ověření byly preferovány zástupci různých taxonomických skupin. 2020 - Czech The methodology of processing hemispherialc photographs is a complex set of activities, starting with the creation of an image with a fish-eye lens, image editing in graphic editors and ending with analysis in specialized software. The obtained data indicate the amount of radiation falling into the undergrowth and the method of processing affects their quality. The procedure developed by the GIS and Remote Sensing Department of BU CAS integrates methods from various programmes into a script in R. Metodika zpracování hemisférických fotografií je komplexní soubor činností, počínaje vyhotovením snímku objektivem typu rybí-oko, úpravou snímku v grafických editorech, končíc analýzou v specializovaném softwaru. Získané údaje vypovídají o množství záření dopadajícího do podrostu a způsob zpracovávání ovlivňuje jejich kvalitu. Postup vyvinutý Oddělením GIS a DPZ BU AV ČR integruje postupy z různých programů do skriptu jazyka R. Detection and identification of aflatoxins by means of portable liquid chromatograph for purpose of control of biological weapons prohibition

2020 - Czech Method describes screening detection and identification of selected mycotoxins aflatoxins (aflatoxins B1, B2, G1 and G2), ochratoxins A, B and zearalenon by means of portable miniaturzed capillary liquid chromatograph (miniLC) with UV/VIS and fluorescence detector (FLD). The aim of the method is to provide rapid screening method for finding/excluding the presence of given mycotoxins in unknown samples. In case of positive finding, it is necessary to perform confirmation analysis using other instrumental methods (e. g. LC-MS). The method is intended namely for the needs of technical support of the State Office for Nuclear Safety for control and supervising activities in field of execution of the Act No. 281/1997 Coll (Biological Act). Metodika popisuje způsob screeningové detekce a identifikace vybraných mykotoxinů aflatoxinů (aflatoxinů B1, B2, G1 a G2), ochratoxinů A, B a zearalenonu pomocí přenosného miniaturizovaného kapalinového chromatografu (miniLC) s UV/VIS a fluorescenčním detektorem (FLD). Cílem metodiky je poskytnout rychlou metodu pro zjištění/vyloučení přítomnosti uvedených mykotoxinů v neznámém vzorku. V případě pozitivního zjištění je nutné u daného vzorku provést potvrzení identity látek pomocí dalších instrumentálních metod (např. LC-MS). Metodika je určena zejména pro potřeby technické podpory SÚJB při kontrolní a dozorové činnosti v oblasti dodržování Zákona č. 281/1997 Sb. 2020 - Czech The presented methodology represents an improvement of the currently used system of wildfire monitoring and forecast. Based on extensive testing, it defines two fire weather indices. The method shows how the combination of both indices can lead to robust fire risk estimates. Both indices were uset to generate a forecast of fire weather, but also to quantify th erelationship between the frequency of favorable conditions for the occurrence of natural fires and their observed numbers, in several time steps (from 1 day to the entire season) The methodology has been implemented within the www. firerisk.cz portal, where it serves as a tool for operational fire risk forecasting. The text of the methodology therefore contains instructions for using the menthodology therefore contains instructions for using the firerick.cz portal. Great attention is paid in the methodology to the aspect of climate change and estimating the impact of this phenomenon on the frequency of wildfires in the Czech republic. Předkládaná metodika představuje vylepšení doposud používaného systému sledování a předpovědi rizika meteorologických podmínek vhodných pro vznik přírodních požárů. Na základě obšírného testování definuje dva indexy požárního počasí, jejichž kombinací lze dospět k robustním odhadům požárního rizika. Oba indexy byly použity pro generování předpovědi požárního počasí, ale také ke kvantifikaci vztahu mezi četností příznivých podmínek pro vznik přírodních požárů a jejich reálného počtu, a to v několika časových krocích (od 1 dne po sezónu). Metodika je nyní rutinně implementována v rámci portálu www.firerisk.cz , kde slouží jako nástroj operativní předpovědi požárního rizika. Text metodiky tak zároveň obsahuje i návod na použití portálu firerisk.cz. Modified method for evaluation of micronuclei in lymphocytes using hybridization of pancentrometric fluorescently labelled probes

2020 - Czech This methodology is based on the simultaneous fluorescent staining of both whole chromosomes and their centromere using pancentrometric probes. Thanks to this, it is possible to evaluate the frequencies of micronuclei with / without centromeres, ie to determine the differences between structural and numerical aberrations. The aim of the methodology was to create an innovative procedure for evaluating the genotoxic effects of nanoparticles, chemicals, including drugs or radiation on the DNA of living organisms. Tato metodika je založena na současném fluorescenčním barvení jak celých chromozomů, tak jejich centromer pomocí pancentrometrických sond. Díky tomu je možné vyhodnotit frekvenci mikrojader s / bez centromer, tedy určit rozdíly mezi strukturálními a numerickými aberacemi. Cílem metodiky bylo vytvořit inovativní postup pro vyhodnocení genotoxických účinků nanočástic, chemických látek včetně léků či záření na DNA živých organismů. 2020 - Czech Metodika předkládá postup (pravidla) pro strukturu metadatového záznamu a popis technických a administrativních metadat z webového archivu. Nastiňuje teoretický rámec problematiky tvorby metadat webových archivů, popisuje granularitu záznamu a jednotlivá metadatová pole s hodnotami, kterých mohou nabývat, a jejich možnou formou zápisu. Vychází především z metadatové specifikace pro webové archivy mezinárodního konsorcia International Internet Preservation Consortium. Metodika byla vyvinuta pro potřeby Českého webového archivu NK ČR (Webarchiv). Je navržena tak, aby mohla sloužit dalším archivům webového obsahu. Jejím účelem je popis technických a administrativních metadat, která se vztahují k jednotlivým sklizním včetně kontejnerového formátu WARC. V souvislosti s touto metodikou byl vyvinut nástroj - software Grainery, který lze použít pro generování, extrakci, evidenci a zobrazení metadat podle postupů stanovených v metodice. 2020 - Czech The ceramic (decorative or utility) accompanies humanity from prehistoric times to the present days. Therefore, the ceramic play important role in collections in all institutions. Due to the nature of the material are those items extremely sensitive to mechanical damage. The submitted method explains the damage mechanism or destruction of the items of cultural heritage. The method also summarizes recommendations for designing the optimal condition of depositing, exhibiting, conservation/restoration, and the manipulation with such sensitive material. Including a description of several methods useful to make quality documentation and replication of cultural heritage items (3D models). Keramika na bázi keramických materiálů (dekorativní i užitková) provází lidstvo od pravěku až po současnost; je proto významnou součástí sbírek všech sbírkotvorných institucí. Vzhledem k povaze materiálu jsou však tyto předměty extrémně náchylné zejména k mechanickému poškození. Předkládaná metodika je souhrnem informací o možných příčinách (mechanismech) poškození či destrukce tohoto typu předmětů kulturního dědictví; zahrnuje však i soubor doporučení pro vytvoření optimálních podmínek deponování, vystavování, konzervování/restaurování a pro manipulaci s předměty, a to včetně popisu několika metod využitelných ke zhotovení kvalitní dokumentace a věrné virtuální replikace předmětů (3D modely). Jar Test Methodology for Water Treatment Optimization

2020 - Czech The methodology is intended for water treatment plant (WTP) operators and serves as a basic guideline for an optimization of the coagulation process in water treatment. It describes the jar test procedure, which allows the determination of the optimum operating conditions of coagulation, i.e., type and dose of the coagulant and coagulation pH value, needed for reaching the highest treatment efficiency. The publication further contains a supplementary part with tables for conversion of molar to mass concentration (and vice versa) of coagulants, instructions for diluting coagulants and reagents for pH adjustment and examples of jar test data recording, processing and evaluation. Metodika je určena provozovatelům úpraven vod a slouží jako podklad pro optimalizaci procesu koagulace při úpravě vody. Podrobně popisuje postup provádění laboratorních koagulačních/flokulačních testů, tzv. sklenicových optimalizačních zkoušek (angl. jar tests), které umožňují určit optimální provozní podmínky koagulace, tedy typ a dávku koagulačního činidla a hodnotu pH, při které musí koagulace probíhat, aby bylo dosaženo co nejvyšší účinnosti úpravy vody. Součástí metodiky jsou přílohy s tabulkami pro přepočet hmotnostních a látkových koncentrací koagulačních činidel, postupy ředění koagulačních činidel a činidel pro předúpravu pH či vzorové tabulky pro záznam a vyhodnocení dat sklenicových optimalizačních zkoušek.
