Protective effect of E. coli strain Nissle 1917 and E. coli strain O83 on the development of inflammation in dextran sulphate sodium-induced colitis in mice

The study was focused on the evaluation of probiotic bacteria E. coli strain Nissle 1917 and E. coli strain O83 and their role in the development of intestinal inflammation in DSS model of ulcerative colitis and the route of the application. Intragastrically tubing SCID mice ( with severe combined immunodeficiency) had heavy chages (inflammation) of the colon – part descendens. Intrarectally application had a protective effect on the development of intestinal inflammation – part colon descendens. Proinflammatory cytokines TNF – alpha and IL – 6 were reduce markedly in fragment culture from colon - descendens Tato práce byla zaměřena na zhodnocení probiotických bakterií E. coli strain Nissle 1917 a E. coli strain O83 .Vývoj střevního zánětu byl studován v DSS modelu ulcerózní kolitidy. SCID myším byly aplikovány probiotické bakterie jak intragastricky, tak intrarektálně. Při intragastrické aplikaci se u experimentálních zvířat objevily těžké zánětlivé změny na sliznici tlustého střeva – část descendens a produkce prozánětlivých cytokinů TNF – alpha a IL – 6, které byly vyizolovány z fragmentových kultur tlustého střeva – části colon – descendens, byla výrazně zvýšena proti skupině u které byly bakterie aplikovány intrarektálně a kde se projevil protektivní vliv aplikovaných bakterií

Recent molecular and functional studies from our laboratory elucidate key aspects in development of porcine T and B lymphocytes including their ontogeny, behavior, fate, subset discrimination, repertoire and specificity of their cell surface receptors. Our results indicate that rather then immune system of swine is unique its understanding may help to elucidate and elaborate some general assumption and hypothesis based by examination of limited number of species such as humans or rodents Současné molekulárně biologické a funkční experimenty naší laboratoře vysvětlují zásadní aspekty vývoje prasečích T a B lymfocytů, jejich ontogenezi, chování, osud, specifičnost, rozdělení, repertoár a zaměření jejich specifických receptorů. Prezentované výsledky prokazují, že spíže než by imunitní systém prasat byl výjimečný, umožňuje nám jeho pochopení pozměnit či upřesnit některá obecná tvrzení a teorie vytvořené na základě studia omezeného počtu zvířecích druhů, jako jsou lidé či hlodavci

Birch pollen is one of the most frequent airborne allergen of Central and North Europe. The effect of intestinal microflora on sensitization and mucosal tolerance to main component of birch pollen Bet v 1 did not be study yet. Germ-free and conventional mice were administrated intragastrically or intranasally with Bet v 1 following by subcutaneous injections of Bet v 1. Tolerance was established independently on microflora present resulting decreased level of IgE and changed cytokine production Březový pyl patří mezi nejrozšířenější vzduchem šířené alergeny střední a severní Evropy. Vliv střevní mikroflóry na senzibilizaci a navození slizniční toleranci k hlavní alergizující komponentě březového pylu - Bet v 1, dosud studován nebyl. Bezmikrobní a konvenční myši byly sondovány Bet v 1 intragastricky a intranasálně a po té byly senzitizovány subkutálně Bet v 1. Byla navozena tolerance nezávisle na přítomnosti bakterií, která se odrazila ve snížené hladině IgE a změněné cytokinové produkci
