2023 - Czech The aim of this study was create three subpopulation of macrohages from human cell lineage THP-1 using different stimulating agents and differentiation factors. Morhologically compare and differenctiate them based on the expresion of surface markers. Furhermore, to perform inflammatory activation using LPS and to test the effect of two selected tick serpins on inflammatory cell activation. Cílem práce bylo vytvořit tři subpopulace makrofágů z lidských liniových THP-1 buněk pomocí různých stimulačních látek a diferenciačních faktorů. Morfologicky je porovnat a rozlišit na základě exprese povrchových markerů. Dále provést zánětlivou aktivaci pomocí LPS a otestovat vliv dvou vybraných klíštěcích serpinů na zánětlivou aktivaci buněk. Jsou osobnostní rysy hraboše polního provázány s denním rytmem pohybové aktivity?

2023 - Czech In this study, the relationship between personality traits and daily activity of Common voles (Microtus arvalis) was investigated. Thirty four voles were captured for tests from the wild and kept separately under stable light (L:D, 12:12) and ambient temperature (19 ? 1 oC) laboratory conditions. Voles were subjected to Open Field (OF) tests to determine their personality traits on locomotor activity and shy-bold gradients. They were then monitored in the PhenoTyper boxes, where their activity was recorded for seven days. In this thesis, it was found that more locomotor active and bold individuals in the OF test are also more active in the PhenoTyper box, are more prone to periodicity and regular ultradian rhythms, and have more activity in the dark. Shy individuals are often immobile (most often in the dark), which weakens the periodicity of their ultradian rhythms. Thus, greater boldness in the common vole leads to greater nocturnal activity, and shyness rather to greater nocturnal immobility. The personality traits were associated with daily activity, periodicity, and ultradian rhythms. V této studii byl zkoumán vztah osobnostních rysů a denní aktivity jedinců hraboše polního (Microtus arvalis). Třicet čtyři hrabošů bylo odchyceno ve volné přírodě a chováno odděleně za stabilních světelných (L:D, 12:12) a teplotních (19 ? 1 oC) laboratorních podmínek. Hraboši byli podrobeni Open Field (OF) testům pro zjištění jejich osobnostních rysů na gradientech pohybové aktivity a odvážnosti. Poté byli podrobeni sledování ve PhenoTyper boxech, kde byla po celých sedm dní zaznamenávána jejich aktivita. V této diplomové práci bylo zjištěno, že více aktivní a odvážní (bold) jedinci v OF testu jsou také více aktivní ve PhenoTyper boxu, mají větší sklony k periodicitě a pravidelným ultradiánním rytmům a mají větší aktivitu ve tmě. Bázliví (shy) jedinci jsou často neaktivní v určitou dobu (nejčastěji v noci), což oslabuje jejich periodicitu ultradiánních rytmů. Tedy větší odvážnost u hraboše polního vede k větší noční aktivitě a bázlivost spíše k větší noční imobilitě. Osobnostní rysy byly spojeny s denní aktivitou, periodicitou a ultradiánními rytmy. It explores the formation and evolution of the subgenre, its influences, and significant characteristics. Additionally, it reflects the specific visual aspects of the grunge style, which both reflected and defined the youth of that time. The work also analyzes the messages and ideas associated with the subculture, often introducing new perspectives on authenticity, freedom, and individuality. It aims to contextualize grunge within a new framework using the theories of sociologists Dick Hebdige and Erich Fromm. Through this analysis, it seeks to understand the elements and values that shape the grunge community and how they manifest in the lives of its members. Through this comprehensive approach, the thesis endeavors to aptly elucidate the multifaceted nature and depth of the grunge subculture, highlighting its significant position within cultural and social contexts. Tato diplomová práce se zaměřuje na hudební subžánr grunge, a především na subkulturu, která k němu náleží. Zkoumá vznik a rozvoj subžánru, jeho vlivy i signifikantní znaky. Reflektuje rovněž specifické vizuální aspekty stylu grunge, které odrážely a jistým způsobem i definovaly tehdejší generaci mladých. Analyzuje také poselství a myšlenky spojené se subkulturou, často přinášející nové pohledy na autenticitu, svobodu a individualitu. Práce usiluje o zasazení grunge do nového rámce s tezemi sociologů Dicka Hebdige a Ericha Fromma. Skrze tuto analýzu se snaží porozumět, jaké prvky a hodnoty grunge komunitu formují a jakým způsobem se promítají do života jejích členů. Tímto komplexním přístupem se práce snaží vhodně osvětlit mnohostrannost a hloubku subkultury grunge a ukázat její významné místo v kulturním a sociálním kontextu. It seeks to offer a view of image as a tool of fashion and ideological dictate, mentioning some significant figures of the decades and their influence on the clothing revolutions. Attention is also paid to rebellious subcultures rejecting mass trends or dark chapters of history, when the forced suppression of image against the background of political events played a role. 2023 - Czech With help of methodological and data triangulation, the online written and oral communication of a virtual project team was investigated in order to answer the question of which strategies the team members develop in multilingual interaction via different media. Light is shed on the use of ELF as a genuinely multilingual practice. The central role of pronounced multilingual competence of the project leaders, as well as the advantages of communication in the mother tongue for building a familiar relationship become evident. The communication in the team presents itself as open to language changes, which allows creativity and joy in the use of language. Mit Hilfe von Methoden- und Datentriangulation wurde die schriftliche und mündliche Online-Kommunikation eines virtuellen Projektteams untersucht, um die Frage zu beantworten, welche Strategien die Teammitglieder in der mehrsprachigen Interaktion über verschiedene Medien entwickeln. Dabei wird der Einsatz von ELF als genuin mehrsprachige Praxis beleuchtet. Die zentrale Rolle einer ausgeprägten mehrsprachigen Kompetenz der Projektleiterinnen und Projektleiter sowie die Vorteile der muttersprachlichen Kommunikation für den Aufbau einer vertrauten Beziehung werden deutlich. Die Kommunikation im Team präsentiert sich als offen für Sprachwechsel, was Kreativität und Freude am Sprachgebrauch ermöglicht. 2023 - Czech The thesis "Tax optimization of employee benefits in a selected company" deals with the issue of employee benefits in the Czech Republic and their tax aspects. The issue of employee benefits is an important part of employee remuneration and serves as an important motivational element. The aim of the thesis is to assess the impact of employee benefits on the employer's income tax. The thesis first focuses on the description and sorting of employee benefits and their legislative regulation in the Czech Republic. The most used benefits are analysed from the point of view of the impact on the income tax of natural and legal persons on the part of the employee and the employer. The described theory is applied to the selected company. Based on the analysis of the current system of benefits provided and the conducted questionnaire survey are suggested changes in the system of benefits with a focus on tax optimization. In conclusion, the impact of these suggestions on company costs and income tax is evaluated. This thesis will be valuable to employers, employees, and tax professionals seeking to reduce tax liabilities and improve the efficiency of employee benefit programs. Diplomová práce se zabývá srovnání výpočtu daně z příjmů mezi osobou samostatně výdělečně činnou a společností s ručením omezeným. Teoretická část práce je zaměřena na vysvětlení pojmů vztahující se k podnikatelské činnosti a formám podnikání fyzických a právnických osob. Po přiblížení hlavních kritérií pro různé formy podnikatelské činnosti je pozornost věnována evidenčním povinnostem uvedených subjektů s cílem stanovit základ daně a vypočítat co nejnižší daň. Cílem praktické části je porovnat, která právní forma podnikání je pro uvedenou osobu v konkrétních podmínkách vhodnější z pohledu daňového zatížení daní z příjmů. Pro jednotlivé případové studie je vypočítána částka daně z příjmů, pojistné na veřejné zdravotní pojištění a sociální zabezpečení současně s doplněním dalších souvisejících skutečností, které se k dané formě podnikatelské činnosti vážou. The Potential of Geocaching as a Tool for Exploring Regions and Landscapes.

2023 - Czech Geocaching is an outdoor activity where participants use GPS technology to find hidden caches. These caches are located using coordinates provided online, and players then embark on a real-world search to locate them. The aim of this diploma thesis is to closely examine the impact of geocaching on landscape exploration within a region. It aims to explani fundamental terms used in geocaching, core gameplay principles, and related concepts for readers. The research section builds on these concepts to addressing predefined research questions and seeks to provide insight into broader context. As part of the recommendations, summary overviews of selected caches are presented. Subjectively selected caches are evaluated based on predefined criteria, and their profiles are provided. Geocaching je venkovní aktivita, při které účastníci využívají GPS technologii k nalezení skrytých schránek. Tyto schránky jsou lokalizovány pomocí souřadnic poskytovaných online. Hráči se poté vydávají přímo do terénu s cílem najít tyto schránky. Cílem diplomové práce je blíže prozkoumat vliv geocachingu na poznávání krajiny v regionu. Vysvětlit čtenářům některé základní pojmy užívané v geocahingu, základních principů hry a dalších souvislostí. Výzkumná část poté plynule navazuje na stanovené výzkumné otázky a snaží se vysvětlit i některé další souvislosti v širším kontextu. V rámci doporučení jsou zpracovány přehledové sumáře některých, subjektivně vybraných keší. Vybraným keším bylo přiděleno subjektivně udělené hodnocení na základě předem stanovených kritérií. 2023 - Czech The aim of this diploma thesis is to present similarities and differences in the structure and dynamics of government expenditure in selected European countries with an emphasis on causal connections. This work examines the function of public finances and the role of the government sector. It further explains the international classification of government spending, and it focuses on the influences that act on the size and structure of government spending. The analytical part of this work includes calculations of government expenditures that are divided into four groups. All the groups are examined in percentage and per capita based on formulas discussed in methodology. Hlavním cílem této práce je najít podobnosti a rozdíly ve struktuře a dynamice vládních výdajů s důrazem na nalezení příčinných souvislostí ve vybraných zemí Evropy. Teoretická část této práce je zaměřena na funkci veřejných financí, úlohu vládního sektoru a mezinárodní klasifikaci vládních výdajů. Dále se zaměřuje na vlivy, které působí na velikost a strukturu vládních výdajů na základě empirických studií. Analytická část této práce obsahuje výpočty vládních výdajů, které jsou rozděleny do čtyř skupin. Všechny skupiny jsou zkoumány v procentech a v přepočtu per capita na základě vzorců diskutovaných v metodice.
