Capillary electrophoresis with contactless conductivity detection for rapid determination of meldonium in urine
Šlampová, Andrea; Kubáň, Pavel
2016 - English
A simple, rapid and cheap capillary electrophoresis-capacitively coupled contactless\nconductivity detection (CE-C4D) method for determination of meldonium (MEL) in\nurine samples was developed. Sample pretreatment was minimized to dilution of urine\nsamples with DI water and MEL was determined by CE-C4D in background electrolyte\nconsisting of 2 M acetic acid (pH 2.3) in ng mL−1 to mg mL−1 concentration range.\nSample-to-sample analysis time was less than 4 min, limit of detection and\nquantification was 0.015 and 0.05 μg mL−1, respectively, and the method showed\nexcellent linearity (r2 ≥ 0.9998), recovery (97.6–99.9%) and intraday as well as interday\nrepeatability (RSD ≤ 4.7%). Keywords: meldonium; capillary electrophoresis-capacitively coupled contactless conductivity detection; urine Available in digital repository of the ASCR

Geochemistry of Cenozoic volcanic rocks of the Faroe Islands with a focus on their ultra-trace contents of Hg
Šišková, P.; Krmíček, Lukáš; Coufalík, Pavel
2016 - English
According to geophysical research performed in the eastern part of the North Atlantic, many authors inferred that the basalt flows on the Faroe Islands overlie a continental metamorphic basement supposedly of Precambrian age. With respect to this, we focused our study on determination of ultratrace mercury contents by means of atomic absorption spectrometry to test this hypothesis. Mercury contents in basalts seem to be a sensitive tool for distinguishing between the basalts not affected by interaction with the continental crust and the basalts enclosing crustal xenoliths. Contents of Hg show great variability between samples even within the same formation. We presume that high concentrations of Hg in some samples are related to stagnation of individual pulses of parent magma in the continental crust below the Faroe Islands. Keywords: Faroe Islands; tholeiitic basalt; mercury content; continental crust Available in digital repository of the ASCR

Concentration of atomic hydrogen measured in a heated atomizer by two-photon absorption laser-induced fluorescence
Dvořák, P.; Talába, M.; Dědina, Jiří; Kratzer, Jan; Obrusník, A.
2016 - English
Concentration of free hydrogen atoms was measured by the two-photon absorption laser-induced fluoresence (TALIF) in an externally heated atomizer in a mixture of argon, hydrogen and oxygen at atmospheric pressure. The atomic hydrogen concentration in the order 10^22 m^-3 was found in the region around the flame. Keywords: TALIF; quartz tube atomizer; hydrogen measurement Available in digital repository of the ASCR

CE-nESI/MS in electrode-free design: narrowing the separation channel to 5 μm
Týčová, Anna; Foret, František
2016 - English
Capillary electrophoresis-nanospray/mass spectrometry (CE-nESI/MS) in electrodefree\narrangement represents a very simple instrumentation for analysis of biomolecules.\nSo far, CE-nESI/MS analyses in this design were limited to 10-75 μm ID of the\nseparation channel. Although the work with narrower dimensions brings several\nchallenges, there is a great potential for better sensitivity, improved separation power\nand lower sample consumption. This work is devoted to some practical aspects of\nexperiments in narrow bore channels and systematic evaluation of CE-nESI/MS\nanalyses conducted in capillaries of 25, 15 and 5 μm ID. Keywords: CE-nESI/MS; electrode-free arragement Available in digital repository of the ASCR

Comparative evaluation of protein composition in human breast cancer cells using mass spectrometry
Flodrová, Dana; Toporová, L.; Macejová, D.; Laštovičková, Markéta; Brtko, J.; Bobálová, Janette
2016 - English
Bottom-up proteomic approach was used for detailed characterization of proteins from\ntwo human tumour cell lines representing major clinically different types of breast\ncancer. The aim was to show the differences between them on proteomic level. Here\nwe present almost 100 unequivocally identified proteins out of which 60 were mutually\ndifferently expressed for MDA-MB-231 and MCF-7. Some well-known breast cancer\nmarkers like annexins A1, A2 and vimentin were found in the MDA-MB-231 cell line.\nOn the other hand, MCF-7 cells were found to be positive for cytokeratins and keratins\nand thus we were able to distinguish both cell lines sufficiently. Keywords: mass spectrometry; breast cancer; human tumour; bottom-up proteomic approach Available in digital repository of the ASCR
