Escherichia coli O83:K24:H31 – a potential live vaccine delivery vector

The aim of our study was to create a mucosal vaccine for impoverished population. We used Escherichia coli O83:K24:H31 as live vector. It produced fused antigen M. tuberculosis fused antigens Ag85B-Esat6 (H1) which was inserted into a surface-exposed loop of the outer membrane protein LamB. Mice were given the bacteria according to the immunization protocol, they received free soluble H1 antigen with the mucosal adjuvant LTK63 as intranasal boost. We observed increase of total IgA on the intestine mucosa after colonization of E. coli O83 and induction of specific anti-H1 IgA level in small intestine Cílem naší práce bylo vytvořit mukosální vakcínu pro třetí svět. Jako vektor jsme použili Escherichia coli O83:K24:H31. Ta byla geneticky manipulována k produkci fůzního antigenu Ag85B-Esat6 (H1) M. tuberculosis, který byl vložen do proteinu LamB. Myši dostaly baktérii podle imunizačního protokolu a byly intranasálně boostovány LTK63 a volným H1 proteinem. Pozorovali jsme zvýšení celkového IgA ve střevě po osazení E. coli O83 a indukci specifických anti-H1 protilátek v tenkém střevě

Yeast Initiation factor 3 (eIF3) occurs together with the eIF2.GTP.Met-tRNAiMet ternary complex (TC) and eIFs 1 and 5 in a pre-formed unit designated the multifactor complex (MFC) that was implicated in playing a critical role in ecient recruitment of TC and mRNA to the 40S ribosomes and stimulation of the post-assembly processes such as scanning and AUG recognition. In ecort to identify binding sites of the MFC on the 40S ribosome, we previously demonstrated that deletion of the rst 199 amino acids of the N-terminal domain (NTD) of the TIF32 subunit of eIF3/eIF3a, in the presence of a wild-type gene, completely eliminated association of the mutant MFC with the 40S ribosome without aecting its overall integrity. We looked for conditions leading to activation of silent gene clusters implied from the sequence as responsible for biosynthesis of yet unknown secondary metabolites in mutants blocked in biosynthesis of known antibiotics (spiramycin, congocidin). This approach should contribute to the better understanding of interaction between primary and secondary metabolism during production of antibiotics and the role of morphological differentiation in these processes U série mutant blokovaných v biosyntetických dráhách známých antibiotik (spiramycin, congocidin) jsme hledali podmínky pro aktivaci přítomných „mlčících“ souborů genů pro biosyntézu zatím nedefinovaných sekundárních metabolitů. Cílem je přispět k lepšímu pochopení principů interakce mezi primárním a sekundárním metabolizmem během produkce antibiotik a objasnit úlohu morfologické diferenciace v těchto procesech
