The aim of the methodology is to present methods for quantifying the impacts of projected climate change on the water balance when applying adaptation measures in the Czech Republic for the next few decades. Adaptation measures should contribute to the sustainability of the water balance in all major user segments of water use and management in the basin. The main principle is the use of hydrological models to transform climate change scenarios into time series of hydrological conditions and to quantify the overall water balance of the basin using different types of adaptation measures and their implementation over time. Special emphasis is placed on the evaluation of combinations of adaptation measures that cannot be analysed by simplified methods. The methodology is designed to search for the optimal combination of adaptation measures in the assessed catchment. The proposed approach eliminates the shortcomings of effectiveness assessment from the perspective of the exclusive user of the water resource, as the evaluation of the effectiveness of adaptation measures is carried out in the form of a multi-criteria analysis of the evaluation of the outputs of the simulation model for predicting the water balance in the whole basin. This methodology can be used to assess different adaptation measures in all basic segments of water users: agriculture, forestry, energy, water management and others. Cílem metodiky je představit metody, jak kvantifikovat dopady předpokládané klimatické změny na vodní bilanci při užití adaptačních opatření v podmínkách ČR na několik příštích dekád. Adaptační opatření mají přispívat k udržitelnosti vodní bilance při ve všech hlavních uživatelských segmentech užití a nakládání s vodami v povodí. Hlavním principem je využití hydrologických modelů, prostřednictvím nichž lze scénáře změny klimatu transformovat do časových řad hydrologických poměrů a kvantifikovat celkovou vodní bilanci povodí s užitím adaptačních opatření různých typů a jejich realizace v čase. Speciální důraz je kladen na hodnocení kombinace adaptačních opatření, které nelze analyzovat zjednodušenými metodami. Metodika je navržena tak, aby bylo možné hledat optimální kombinaci adaptačních opatření v posuzovaném povodí. Navrhovaný postup eliminuje nedostatky hodnocení účinnosti z pohledu výlučného uživatele vodního zdroje, protože vyhodnocení účinnosti adaptačních opatření je realizováno formou multikriteriální analýzy vyhodnocení výstupů simulačního modelu predikce vodní bilance v celém povodí. Touto metodikou lze posuzovat různá adaptační opatření ve všech základních segmentech uživatelů vody: zemědělský, lesnický, energetický, vodohospodářský a další. Complex survey of the fish community of the Milada Lake in 2022

The report summarizes the results of a comprehensive survey of the fish population of Lake Milada in 2022. The goal of the ichthyological survey of the lake was to map the state and development of the fish population, with an emphasis on estimating the abundance and biomass of individual fish species, on estimating the risks of further development of the fish population and recommending further management of the lake. A summary of the achieved results and a proposal for recommendations for further monitoring are the contents of the presented report. Zpráva shrnuje výsledky komplexního průzkumu rybí obsádky jezera Milada v roce 2022. Cílem ichtyologického průzkumu jezera bylo zmapování stavu a vývoje rybí obsádky zejména s důrazem na odhad početnosti a biomasy jednotlivých druhů ryb, na odhad rizik dalšího vývoje rybí obsádky a doporučení dalšího obhospodařování jezera. Structural analysis of polysaccharides from mushroom hericium Erinaceus mftccb 134

The aim of this research was the structural characterisation of polysaccharides isolated from fruiting bodies of the cultivating medicinal mushroom Hericium erinaceus MFTCCB 134. Cold and hot water extracts (CWE, HWE) were obtained and purified, and followed by alkali extraction from the reminder solids. The composition and structure of the fractions were analysed by FTIR spectroscopy, and neutral sugars/linkage analyses were determined by GC/FID and GC/MS, respectively. Galactose, fucose and glucose were found in purified CWE,\nwhile glucose predominated in purified HWE. Analytical Derivatization for Identification of Electrochemical Oxidation Products of Fentanyls

Analytical derivatization and GC-MS analysis were employed for the identification of main\nproducts from the electrochemical oxidation of fentanyl derivatives. Chemical derivatization\nprovides proof of the chemical reactivity of functional groups in oxidation products and serves\nfor the chemical conversion of target compounds into the second series of derivatives to confirm\nthe identification of particular products. Using the direct GC-MS analysis of oxidation products,\nderivatization of aldehydes with cysteamine and derivatization of amides by acidic hydrolysis\nand subsequent reaction with ethylchloroformiate, phenylacetaldehyde, and Nphenylpropanamide\nwere unambigously identified as oxidation products from fentanyl,\nthiophen-2-ylacetaldehyde, and N-phenylpropanamide from sufentanil, and\nphenylacetaldehyde and 2-furanilid from furanylfentanyl. Analytická derivatizace a plynová chromatografie s hmotnostní spektrometrií se osvědčily jako\nvhodné nástroje pro identifikaci oxidačních produktů a řešení reakčního mechanismu\nelektrochemické oxidace fentanylů a dalších opioidů. Provedené experimenty potvrdily\npřítomnost fenylacetaldehydu a N-fenylpropanamidu v roztoku po elektrochemické oxidaci\nfentanylu. V případě sufentanilu byl jako odpovídající aldehydický produkt prokázán thiofen-\n2-ylacetaldehyd a v případě furanylfentanylu byl jako amidický produkt prokázán zcela\nanalogicky 2-furanilid.
