Antibiotics-modes of action, resistance mechanisms and the search for new compounds

According to their mechanism of action antibiotics can be divided into 4 groups. 1. Antibiotics inhibiting bacterial cell wall biosynthesis. 2. Antibiotics inhibiting DNA synthesis. 3. Antibiotics inhibiting RNA synthesis and 4. Antibiotics inhibiting protein synthesis. Basic mechanisms of antibiotic resistance include cell impermeability, modification of target site, modification or degradation of the antibiotic and increased efflux of the antibiotic. The following can be used in the search for new compounds: 1. Microorganisms isolated from extreme life conditions 2. Environmental DNA (eDNA) in which genes coding for secondary metabolites are looked for. 3. Streptomyces genomics yielding information about genes coding biosynthesis of new compounds. 4. Combinatorial biosynthesis for the production of hybrid antibiotics. Podle mechanizmu účinku se antibiotika dělí do 4 skupin: 1. Antibiotika inhibující biosyntézu bakteriální buněčné stěny, 2. Antibiotika inhibující syntézu DNA. 3. Antibiotika inhibující syntézu RNA. 4. Antibiotika inhibující protesyntézu. Základní mechanizmy rezistence vůči antibiotikům jsou: 1. Neprůchodnost buněčných obalů. 2. Modifikace zásahového místa. 3. Modifikace nebo degradace antibiotika. 4. Zvýšený eflux antibiotika. Jako zdroj nových antibiotik mohou sloužit: 1. Mikroorganizmy izolované v extrémních podmínkách prostředí. 2. DNA izolované z prostředí (eDNA), v které jsou hledány geny kódující sekundární metabolity. 3. Genomika streptomycetů poskytující informace o genech kódujících biosyntézu nových látek. 4. Znalosti o biosyntéze sekundárních metabolitů využívané při přípravě hybridních antibiotik. Decomposition of cellulose and hemicelluloses by wood-rotting fungi and effect of heavy metals

Degradation of cellulose and hemicelluloses by brown-rot fungi is significantly affected by heavy metals. The activity of cellulases is increased in the presence of zinc. Rozklad celulózy a hemicelulóz je u hub hnědé hniloby výrazně ovlivněn těžkými kovy. V přítomnosti zinku je zvýšena aktivita celulolytických enzymů. Activities of both cellulolytic and ligninolytic enzymes and heavy metal content in fruit body of Fomes fomentarius were studied. The activities of ligninolytic enzymes were higher in the core. Byly měřeny aktivity ligninolytických a celulolytických enzymů v plodnici F. fomentarius. Současně byly měřeny i obsahy těžkých kovů. Bioavailability of PAH and its effects on biodegradation during composting

The publication deals with application of composting process on remediation of soils contaminated with PAHs. Bioavailability of PAHs was tested with sequential SFE and the data were compared with the results of remediation. SFE is suitable to predict results of bioremediation. Publikace se zabývá aplikací kompostovacího procesu na remediaci půd kontaminovaných PAU. Biodostupnost PAU byla sledována pomocí SFE a tyto data byly korelovány s výsledky remediace.
