Cathodoluminescence imaging of carotenoids in biological specimens using cryo-scanning electron microscopy using a functional sample of CRYTUR cathodoluminescence detector

Here, we report for the first time the cathodoluminescence properties of carotenoids present in different biological specimens (Diplonemidae spp, carrot root, Arabidopsis embryos, algae Vischeria sp., unicellular pigmented yeast Rhodontorula, hen´s egg yolk) using cryo-SEM. The presence of carotenoids was proven by Raman spectroscopy. Zde poprvé referujeme o katodoluminiscenčních vlastnostech karotenoidů přítomných v různých biologických vzorcích (Diplonemidae spp, kořen mrkve, embrya Arabidopsis, řasa Vischeria sp., jednobuněčná pigmentovaná kvasinka Rhodontorula, žloutek slepičího) pomocí kryo-SEM. Přítomnost karotenoidů byla prokázána Ramanovou spektroskopií.

Sediment management in reservoirs – methods of sediment handling, sediment reevaluation to product - stabilized solidificate

As part of the research work, a semi-operation was set up and a test polygon (''island'') was built on central Novomlýnská reservoir (Věstonická reservoir). The sediment excavation and its transformation into different type of ''products'' in a special mixing plant. The stabilised solidified product was subsequently used in the construction of the new island. The new island will be used this year for monitoring, including, among other things, the behaviour of the new sediment structural elements, its shape stability and other parameters. As part of the protection of the island from wave action, also from solidified sediment, experimental constructions of submerged breakwaters have also been created. Another way of using the stabilised solidified sediment is outside the reservoir, for example in earthworks and to create a stock of ''soil'' for other construction activities. V rámci výzkumných prací byl zřízen poloprovoz a postaven zkušební polygon (''ostrov'') na střední Novomlýnské nádrži (Věstonická nádrž). Zde probíhala i těžba sedimentu a jeho přetvoření na různé typy ''výrobků'' ve speciálním míchacím zařízení. Stabilizovaný solidifikát byl následně použit do konstrukce nového ostrova. Na novém ostrově bude v letošním roce probíhat monitoring, zahrnující mimo jiné chování nových konstrukčních prvků ze sedimentu, jeho tvarová stálost a jiné parametry. V rámci ochrany ostrova před účinkem vln byly, rovněž ze solidifikovaného sedimentu, vytvořeny i experimentální konstrukce ponořených vlnolamů. Dalším způsobem využití stabilizovaného solidifikátu je využití vně nádrže, například do zemních těles a vytvoření zásoby ''zeminy'' pro jiné stavební akce.
