Práce se zaměří na vyprávěcí perspektivu současného románu britského autora Iana McEwana Nutshell, který autor pojal jako literární experiment ve vztahu k literárnímu odkazu Williama Shakespeara a jeho klasické tragédie Hamlet. Práce v úvodní části představí autora v kontextu současné britské prózy a stručně zmíní díla současné literatury, která jsou součástí projektu The Hogarth Shakespeare Series (od roku 2015). Jádrem práce bude literární analýza McEwanova románu ve vztahu k tragédii W. Shakespeara, která se soustředí na identitu vypravěče příběhu, jeho existenciální zkušenost, vnímání světa a uzavřeného prostoru v matčině lůně. Závěrečná kapitola zhodnotí téma románu I. McEwana v porovnání s námětem klasické Shakespearovy tragédie. The aim of this thesis is to analyse the narrative perspective in Ian McEwan's contemporary novel called Nutshell. The author approached the novel as a literary experiment related to the legacy of William Shakespeare and his classical tragedy Hamlet. Firstly, the thesis presents the author in the context of contemporary British fiction and briefly introduces the works included in a project called The Hogarth Shakespeare Series (since 2015). The core of this thesis is a literary analysis of McEwan's novel in relation to Shakespeare's tragedy. The novel focuses on the narrator's identity, his existential experience, his perception of the world and the closed space of his mother's womb. The last chapter then views the themes included in McEwan's Nutshell in comparison to the theme of a classical Shakespearean tragedy. Plné texty jsou dostupné v digitálním repozitáři JČU.

Předmětem zkoumání bakalářské práce je proměna gotického žánru a jeho parodií v literatuře pro dětské čtenáře. Práce se v teoretické rovině zaměří na charakteristiku gotického žánru v průběhu staletí s ohledem na dynamiku gotických prvků v dílech dětské literatury a poté v interpretační rovině přejde k literární analýze textu románu pro dětské čtenáře The Graveyard Book současného britského autora Neila Gaimana. Práce si klade za cíl porovnání a literární analýzu textu The Graveyard Book s textem parodie gotického románu Oscara Wildea The Canterville Ghost a pokusí se zhodnotit přínos a oblibu gotického žánru v současné britské próze. The subject of the bachelor thesis is the transformation of the Gothic genre and its parodies in literature for children's readers. On the theoretical level, the thesis will focus on the characteristics of the Gothic genre over the centuries with regard to the dynamics of Gothic elements in works of children's literature and then, on the interpretive level, will proceed to a literary analysis of the text of the children's novel The Graveyard Book by the contemporary British author Neil Gaiman. The thesis aims to compare and literary analyse the text of The Graveyard Book with the text of Oscar Wilde's parody gothic novel The Canterville Ghost and attempts to assess the contribution and popularity of the gothic genre in contemporary British prose. Measuring authenticity of dialogues: English textbooks and sitcom scripts

Tato diplomová práce zjišťuje míru autentičnosti modelových dialogů v učebnicích anglického jazyka a scénářů situačních komedií. Jejím cílem je prokázat, zda scénáristé situačních komedií předkládají autentičtější dialogy než autoři učebnic anglického jazyka. Teoretická část je věnována procesu komunikace, jejím modelům a kanálům. Dále je ve stručnosti popsán pragmatický aspekt sociální interakce a je vyzdvižena klíčová role kontextu při procesu komunikace. Pozornost je také věnovaná kolokacím a žánru situační komedie. V praktické části je představena analýza nasbíraných dialogů ze čtyř situačních komedií a učebnic angličtiny pro pokročilé uživatele (úroveň C1-C2). Jsou zvoleny dva sitcomy od britských tvůrců a dva od amerických, které jsou srovnány s britskými a americkými učebnicemi angličtiny (CAE a TOEFL). Výzkum se soustředí na nejčastěji se vyskytující podstatná jména a jejich kolokace. Výsledky výzkumu jsou následně také porovnány s národními korpusy anglické a americké mluvené angličtiny. Výsledek diplomové práce ukazuje, že situační komedie i učebnice pro pokročilé uživatele angličtiny zahrnují autentické dialogy. This diploma thesis measures the authenticity of model dialogues in English textbooks for advanced learners and conversations occurring in situation comedies. The theoretical part focuses on the communication process, its models and channels. Subsequently, context is presented as well as its importance in speech. Attention is then paid to collocations as well as to classification of the genre of situation comedy. The practical part presents the research of sitcom and textbooks dialogues. The data were collected from four sitcoms (two of British and American origin) and CAE and TOEFL textbooks. The analysis concentrates on the most frequent nouns and their collocations. The findings are then compared, and the most frequent expressions are studied with the help of national corpora of British and American spoken English. The result of this thesis is that both textbook and sitcom dialogues include authentic material. Plné texty jsou dostupné v digitálním repozitáři JČU.

Mannan-BAM, TLR ligands, and anti-CD40 immunotherapy in established murine pancreatic adenocarcinoma: understanding therapeutic potentials and limitations

The aim of this RNDr. Thesis is focused on understanding therapeutic potentials and limitations of the antitumor MBTA immunotherapy which is based on synergy of TLR agonists, anti-CD40, and phagocytosis stimulating ligands anchored into the tumor cell membranes. In this study, immunotherapy was tested in murine pancreatic adenocarcinoma Panc02 model. Firstly, the short-term and long-term efficacy of MBTA therapy was tested using established subcutaneous Panc02 tumors two times larger than in previous study. Secondly, the work is devoted to better understanding of the adaptive immunity involvement focusing on CD4+ and CD8+ T lymphocytes during the therapy and their effect on tumor volume reduction, long-term survival and resistance against tumor rechallenge. Subsequently, the ability of immunological memory to cross over the blood-brain barrier confirming its potential applicability in metastatic brain tumors was examined. Moreover, the antigen specificity of the immunological memory was evaluated. Finally, the potential of MBTA therapy to cure metastatic disease, represented by bilateral Panc02 mouse model, was studied. In this case, the MBTA therapy manifested a lower therapeutic response. Therefore, it was combined with diverse therapeutic approaches, such as intratumoral application of anti-CTLA-4 antibody, heat-killed Listeria monocytogenes, chemoablation using EtOH, targeting the tumor microenvironment by hyaluronidase, simultaneous injections of MBTA therapy in primary and secondary distant tumors, and its combination with RT. Despite all these combinations, our results showed that only simultaneous application of MBTA therapy into both tumors has potential for the treatment of the bilateral Panc02. Determinants of Piper (Piperaceae) climber composition in a lowland tropical rainforest in New Guinea

The study examines the distribution of root climbers from the genus Piper on host trees depending on environmental properties and host tree characteristics in a lowland primary tropical rainforest in Papua New Guinea. More specifically, the study examines the relationship between the climbers' presence and various factors including topography (e.g. slope, altitude), vegetation properties, and host tree characteristics. The findings reveal that Piper climbers tend to avoid higher altitudes, steeper slopes, closed canopy layers, and areas with larger total basal area of host vegetation. The preferences of Piper climbers for certain tree species are primarily influenced by the properties of the host trees, particularly their diameter at breast height (DBH). Additionally, the study suggests that Piper species are more commonly found on rare tree species, possibly due to their preference for larger host tree DBH. The results highlight the non-random association between climbers and their host trees, which is influenced by local environmental conditions. These interactions are likely to have important implications for forest vegetation dynamics and the maintenance of diversity. Mechanisms behind diversity and ecosystem functioning relationships - the insights from observational and manipulative studies in grassland communities

This thesis focuses on the relationship between diversity and two commonly used ecosystem functions - productivity and its stability. The thesis tries to assess the problematics from different perspectives using large-scale observational studies and small-scale manipulative experiments to better understand which mechanism create observed patterns. High emphasis is put on the methodological aspects and potential theoretical or practical issues arising from experimental setting and data collection. More specifically, the thesis starts with description and critical evaluation of patterns observed at bigger spatial scales and across multiple sites (Chapter I). Then continue with detailed small-scale experimental approaches and assessment of mechanism driving relationship between diversity, productivity and stability of communities (Chapter II and III). Further, the thesis digs into bigger detail focusing on which plant characteristics define abundance patterns of individual species, which translates into productivity (Chapter IV). Finally, it elaborates on common issues related to vegetation data collection and their subsequent analysis (Chapter V). Cell segmentation from wide-field light microscopy images using CNNs

Image object segmentation allows localising the region of interest in the image (ROI) and separating the foreground from the background. Cell detection and segmentation are the primary and critical steps in microscopy image analysis. Analysing microscopy images allows us to extract vital information about the cells, including their morphology, size, and life cycle. On the other hand, living cell segmentation is challenging due to the complexity of these datasets. This research focused on developing Artificial Intelligence/Machine Learning methods of single- and multi-class segmentation of living cells. For this study, the Negroid cervical epithelioid carcinoma HeLa line was chosen as the oldest, immortal, and most widely used model cell line. Several time-lapse image series of living HeLa cells were captured using a high-resolved wide-field transmitted/reflected light microscope (custom-made for the Institute of Complex System, Nové Hrady, Czech Republic) to observe micro-objects and cells. Employing a telecentric objective with a high-resolution camera with a large sensor size allows us to achieve a high level of detail and sharper borders in large microscopy images. The collected time-lapse images were calibrated and denoised in the pre-processing step. The data sets collected under the transmission microscope setup were analyzed using a simple U-Net, Attention U-Net, and Residual Attention U-Net to achieve the best single-class semantic segmentation result. The data sets collected under the reflection microscope setup were analyzed using hybrid U-Net methods, including Vgg19-Unet, Inception-Unet, and ResNet34-Unet, to achieve the most precise multi-class segmentation result. Detection of Diabetic Retinopathy using Deep Learning and Transfer Learning Techniques with Oversampling to Address Imbalanced Dataset

The study aims to develop a system for detecting diabetic retinopathy using deep learning. In this study I have explored transfer learning with four distinct models and addressed the issue of an unbalanced dataset with oversampling. The final experiment achieved a significant improvement in accuracy and quadratic kappa score. The study highlights the potential of deep learning and the importance of addressing dataset imbalances for accurate results. Vampirism and Queerness in Nineteenth-Century Literature

Cílem této práce je analýza spojení mezi vampirismem a queer motivy v dílech Drákula od Brama Stokera a Krev upírky od Florence Marryat. Diplomová práce je rozdělena do tří částí. První část se zabývá vztahem viktoriánské Anglie k sexualitě a spojitostí mezi queer motivy a gotickým románem 19. století. Věnujeme se zde rozdílnému vnímání mužské a ženské homosexuality a zobrazování tzv. "Others" v dobové literatuře. Ve druhé části se zaměříme na interpretaci díla Drákula z hlediska queer teorie, a to prostřednictvím hraběte Drákuly a role, kterou v románu představuje, včetně jeho vztahu k ostatním mužským postavám. Třetí část se soustředí na analýzu díla Krev upírky, které popisuje osud ženské upírky Harriet Brandt. Nakonec se práce zabývá aspekty jinakosti, kterými Harriet ohrožuje viktoriánskou společnost, a zároveň dvojznačným chápáním tohoto díla. The aim of this thesis is the analysis of the connection between vampirism and queerness in Bram Stoker's Dracula and Florence Marryat's The Blood of the Vampire. The Master's thesis is divided into three parts. The first part examines the relationship of Victorian England to sexuality and the connection between queerness and the 19th-century Gothic novel. We will pay attention to the different perceptions of male and female homosexuality and the portrayal of the so-called "Others" in contemporary literature. In the second part, we will focus on the interpretation of Dracula in terms of queerness, namely through Count Dracula and the role he plays in the novel, including his relations to the other male characters in the novel. The third part concentrates on the analysis of The Blood of the Vampire, describing the fate of female vampire Harriet Brandt. Finally, the thesis explores the aspects of Harriet's Otherness that Harriet threatens Victorian society with, as well as the ambiguous understanding of this work.
