2023 - English
Currently, the nursery production of plant material emphasizes the production of healthy, virus-free material. For this reason, it is necessary to have reference plant materials in which the virus is present. Some types of viruses are difficult to maintain outside the plant host. Because of the potential transmission to outdoor plantings, it is necessary to keep infected plants in special cultivation areas, and net-houses, which brings considerable financial and labour costs. Therefore, a methodology was developed in which the virus status in plant material is analysed, the infected material can be stored using cryopreservation in liquid nitrogen and then regenerated in vitro, and relevant data in GeneEver cryobox can be stored. Keywords: genetic resources; virus; Rubus idaeus L. Fulltext is available at external website.

Detailed Investigation of Human Galectins -1 and -3 by employing selectively deoxyfluorinated N-acetyllactosamines
Kurfiřt, Martin
2023 - English
This work is focused on a detailed investigation of supramolecular recognition events between human galectins -1 and -3 and monodeoxyfluorinated N-acetyllactosamine probes, which were previously prepared in our laboratory. The recognition was studied using a combination of X-ray crystallography, isothermal titration calorimetry and several advanced NMR techniques, such as 19F T2-filter, 1H–1H STD, 1H–15N CSP or 19F EXSY. These techniques provided insight into the thermodynamics and kinetics of recognition and enabled us to uncover of subtle differences between both investigated galectins. Such differences could be potentially exploited in the selective targeting of individual galectins with important implications for the galectins-focused\npharmaceutic industry. Keywords: galectins; NMR techniques; pharmaceutic industry Available in a digital repository NRGL

The electrodiffusional theory for wall shear stress measurement by a two-strip probe: a journey to near-wall region hydrodynamics
Harrandt, Václav
2023 - English
This contribution deals with a new theory describing mass transport in the vicinity of the measuring two-segment strip probe. Analytical formulas for the mass transfer coefficients of the front and the rear electrodes were derived. At the same time, the correctness of these formulas was confirmed by the numerical solution of the convectiondiffusion transport equation. Keywords: transport phenomena; electrodes; wall shear stress Available in a digital repository NRGL

(NE)ZÁVISLOST AKTIVACE AEROSOLU NA POZICI V OBLAKU
Zíková, Naděžda; Pokorná, Petra; Sedlák, Pavel; Sokol, Zbyněk; Ždímal, Vladimír
2023 - Czech
Five in situ campaigns focused on aerosol-cloud interactions were conducted at Mount Milešovka in the Czech Republic to gain more insight into aerosol activation and its dependence on meteorological parameters, mainly vertical air velocity and position within the cloud. The activated fraction was calculated from the difference of concentrations measured behind the whole air inlet and the PM2.5 inlet. The liquid water content (LWC) was calculated from visibility, cloud base position was estimated from ceilometer data. Vertical air velocity was estimated from cloud radar. No strong dependence was found between visibility and vertical velocity, suggesting that the clouds at the station are mostly of advection or inversion origin. Both visibility and LWC depend on the position within the cloud, with the highest LWC values found when the station was between 100 and 400 m above the cloud base, independently of the actual value. 3D Printed Microfluidic Cell for Spectroelectrochemical Analysis
Šikula, M.; Vaněčková, Eva; Hromadová, Magdaléna; Kolivoška, Viliam
2023 - English
Recent advances in fused deposition modeling 3D printing (FDM 3DP) enabled \nthe manufacture of customized spectroelectrochemical (SEC) devices. Despite significant progress, \nreported designs still rely on conventional optical components (windows and cuvettes). In this \nwork, we apply bi-material FDM 3DP combining electrically conductive and optically \ntranslucent filaments to manufacture a fully integrated microfluidic SEC device. Employing cyclic \nvoltammetric measurements with redox probes, we demonstrate that the platform allows SEC sensing of \nreactants, intermediates, and products of charge transfer reactions in an oxygen- free \nenvironment. Developed approaches pave the way for SEC d vices with dramatically\nreduced costs compared to currently available commercial platforms.\n Keywords: spectroelectrochemistry; three-dimensional printing; fused deposition modeling Available on request at various institutes of the ASCR

Electrochemical Properties of Mixed Self-Assembled Monolayers Based on Terpyridine Transition Metal Complexes
Kocábová, Jana; Nováková Lachmanová, Štěpánka; Vavrek, František; Valášek, M.; Hromadová, Magdaléna
2023 - English
This work presents the electrochemical characterization of mixed self-assembled monolayers\n(SAMs) of [Os(terpy)2]2+ and terpyridine connected by tripodal anchoring groups to the\npolycrystalline gold bead electrode and provides a comparison with SAM based on\n[Os(terpy)2]2+ tripodal molecules without diluting terpyridine tripodal molecules. It is shown\nthat both arrangements enable reproducible preparation of well-defined compact films. The\nelectron transfer rate constant for mixed SAM was determined from cyclic voltammograms\naccording to Laviron’s analysis and compared with the rate constant obtained previously for\nSAM containing only [Os(terpy)2]2+ tripod. 2023 - English
Elimination voltammetry with linear scan (EVLS) represents an almost traditional\nmathematical method that helps to obtain additional and detailed information about the recorded\nelectrochemical signals and therefore, to better understand the analyzed systems. Although it\nhas been substantially developed and expanded during its existence of more than a quarter of a\ncentury and has found many areas of application, like any other mathematical and experimental\nmethod, it exhibits several advantages and disadvantages. In this contribution, we summarize\nits strengths, weaknesses, opportunities, as well as threats.\n Keywords: elimination voltammetry with linear scan; advantages; disadvantages Available on request at various institutes of the ASCR
